E621 is largely a porn art site. If you’ve ever seen a “tastefully cropped” image on imgur that someone lists a string of numbers as the sauce, it’s from E621. The comic is saying that Judy Hopps and Twilight Sparkle are fairly close in tagged post numbers. Derpibooru is an mlp-based image sharing site, that has about 10x as many Princess Celestia tags.

... and as for safe vs otherwise, cutting it down to only images marked as explicit or questionable (which probably is as close as a quick check will get to the contents of e621) brings twilight down to about 29k on derpi
